Small-molecule estrogen-related receptor agonist

What is SLU-PP-332?

SLU-PP-332 is a chemical tool targeting the estrogen-related receptors ERRα, ERRβ and ERRγ. Despite their names, these nuclear receptors are distinct from the classical estrogen receptors. SLU-PP-332 is a small molecule rather than a peptide or a naturally occurring exercise hormone.

Mechanisms under investigation

ERRs regulate gene programs associated with energy metabolism. Discovery experiments found activity across all three subtypes, with greatest potency at ERRα in the reported system. The compound changed mitochondrial and respiratory measurements in a muscle cell line and activated an exercise-associated transcriptional program in mice.

Documented lines of inquiry

Research areas

  1. 01

    Muscle-cell bioenergetics

The discovery study examined receptor activation and mitochondrial function in cultured skeletal-muscle cells. Such assays test a molecular hypothesis under controlled conditions. Increased cellular respiration alone does not demonstrate improved health or performance in a whole organism. [1]

  2. 02

    Exercise-associated adaptations

In mice, investigators reported changes in oxidative muscle-fiber characteristics and exercise endurance. ERRα activity was important for the measured response. Calling the compound an exercise mimetic describes selected experimental effects, not replacement of the many physiological consequences of physical activity. [1]

  3. 03

    Obesity and metabolic models

A follow-up study examined diet-induced obese and genetically obese mice. Reported outcomes included increased energy expenditure and fat oxidation, reduced fat accumulation and improved insulin sensitivity. These are model-specific animal findings rather than human weight-management data. [2]

Reading the evidence

What the studies
can tell us.

Cell and mouse evidence

The cited primary publications provide mechanistic and organism-level preclinical findings. Neither is a human intervention trial. They establish a research rationale for studying ERR activation while leaving the translation from laboratory models to human physiology unresolved.

What remains uncertain

Species differences, exposure, off-target effects and prolonged receptor activation require separate investigation. An endurance result in mice does not establish cardiovascular benefit, safety or human athletic effects. Findings should not be transferred to other ERR agonists, and the literature does not validate WYZ material identity or equivalence.
